If you were using, say, nginx, you would just set all requests on the "old" EC2 instance to proxy_pass to the "new" EC2 instance's IP/port.  Any requests that happen to land on the "old" instance would simply be forwarded onto the "new" instance, which would process the request, send back the response and the "old" instance would serve the response back to the requestor.
